Is it possible that we can assign Office 365 License to an employee using API through SAP Integration Suite?

Yes. Have a look at the Microsoft Graph API to achieve this. I believe assignLicense is what you are looking for with CPI. Note, there are blogs out there implementing similar use cases for further inspiration. Have a look at this example with Powershell for instance.
