When working with Personas we see we can access the flavors via both:
https://yyy.co.uk/sap/bc/gui/sap/its/webgui?
And:
https://yyy.co.uk/sap/bc/personas?
Can either URL be used for Personas development and user access?

The regular webgui service call should work equally well. Try removing the question mark at the end of the URL, or simply start transaction WEBGUI which launches SAP GUI for HTML and check the proper URL that way.
The main reason for using the personas service originates from the time when it was necessary to pre-configure webgui for Personas with certain parameters for proper operation. This is mostly needed for older basis releases still. On a more modern basis system, this is not really needed anymore and launching webgui is just fine. In fact, in a public cloud instance, there is no separate personas service available and the only way to access Personas is via using the webgui service.

Can either URL be used for Personas development and user access? Yes, both of them could work.
Just a little bit more clarification ... Both the URL is for launching the SAPGUI for HTML (aka WebGUI). But the 2nd URL is more specific (it has specific instruction to use client 300; with English as login language). Both of this is an equivalent to the root node of SAPGUI for Windows (aka Tcode: SMEN); from which you could go any transaction that you like (SE38, VA01, VA03); only restricted by your security profile.
