I want to know how Personalization segmentation works in OCC V2 API calls in SAP Commerce OCC APIs.
Which API call is responsible to segment the user or invoke personalisation services to update user segment strategy in OCC API call?

Personalization in SAP Commerce Cloud, for the most part, does not use OCC V2 APIs. Personalization relies on its own set of web service extensions along with integrating into the existing cmswebservices extension.
Unless you are using SAP Commerce Cloud, Context-Driven Services | SAP Help Portal - users will need to be assigned to user groups manually or via a third party integration.
For segments:
